A 2010 update of the original NBA Jam, this version retains the classic 2-on-2 gameplay with loose rules and includes all 30 teams from the 2010-2011 NBA season. It introduces a new Remix Mode, adding extra arcade elements to the action.

Players can steal the ball, shove opponents without penalty, use a turbo modifier to boost speed and moves, and go "on fire" after three consecutive baskets. It features elements from later Midway basketball games, such as crossovers, spins, and tag-team alley-oops from NBA Hang Time, as well as extended rosters from NBA Jam Tournament Edition, allowing each team to have at least three players with varied strengths and stats. Injuries are not present, and teams can be expanded with unlockable players earned through in-game achievements.

Remix Mode also brings back power-up icons from NBA Jam Tournament Edition, offering temporary benefits like instant "on fire" or invulnerability to shoves. Other modes include "hot spots" for point bonuses, an "elimination" mode where the lowest scorer each quarter is ejected, a mode where teams try to shatter their opponent’s backboard first, and a variation of the classic street ball game 21.

Players can choose between a Classic campaign, where they face all 29 other NBA teams, or a Remix campaign, where strong performance earns points to unlock new divisions and boss battles against basketball greats. Completing these challenges unlocks new teams and secret cheats. The game features digital photos of players on animated bodies with changing expressions and includes a photo of the team's current coach.
